Effect of Moderate Supplementation with L-Arginine on the Preservation of Trabecular Bone in Long Evans Rats That Have Undergone Ovariectomy

Abstract

With the aim of preventing trabecular bone loss in young Long Evans rats that have undergone bilateral ovariectomy (OVX), we administered L-arginine (L-Arg), a precursor of nitric oxide. Twenty-four rats weighing 250–300 g were divided into three uniform groups: sham OVX (SIM), control OVX, and OVX treated with L-Arg (OVX L-Arg).


Over a 28-day period, the treated group received a 6% (w/v) dose of L-Arg in their drinking water, equivalent to 1.74 g/day, while the OVX and SIM groups received plain water as a vehicle. Biochemical tests (serum alkaline phosphatase, urinary deoxypyridinoline, and NOx), histological analyses of the tibia (density, trabecular bone thickness, and bone marrow), and biomechanical tests of the femur (stiffness, resistance, strength, and maximum load) showed no significant changes. Conclusion: The dose of L-Arg administered for 28 days in drinking water did not preserve the integrity of the trabecular bone in OVX L-Arg rats.
